ASSESSMENT OF HEXANE IMPACT UPON THE HEMATOPOIETIC SYSTEM OF RATS IN ACUTE EXPERIMENT |

Today, the question of impact of aliphatic hydrocarbons upon human and animal organisms is highly topical. The major component specifying their effect is hexane. It is evident that hexane is a polytropic poison affecting all kinds of body tissues. On the basis of the foregoing, the research objective is to study the nature of hexane impact upon the hematopoietic system of rats in the conditions of acute experiment.

The
mature male rats of 170-210 g each were exposed to hexane in the dose of 1/3
CL50 in standard 200 liter Kurlyandskiy chambers. The experimental
animas were divided in 4 groups: the st one was the reference group:
intact animals that were on general vivarium feed and not exposed to hexane;
the 2nd group: animals that were exposed to hexane in the dose of
1/3 CL50 within 4 hours and killed immediately after the inhalation;
the 3rd group: animals that were exposed to hexane at the same time
with the 2nd group and killed in three hours after the inhalation;
and the th group which was also exposed to hexane at the same time
with the 2nd group and killed in 12 hours after the inhalation. After
the expiration of hexane impact period the rats were killed with instant decapitation.
The condition of lipids microsomal oxidation in spleen was studied by the level
of products reacting with thiobarbituric acid (TBA-RP). The level of diene conjugates
(DC) was assesses by the extinction level of washed homogenates at the light
wavelength of 233 nm. The condition of antioxidant protection (AOP) was assessed
by catalase activity. To determine the Cholesterin level in the tissues there
was used the lipids extraction method proposed by Folch and modified by Uzbekov
V.A. and Yagufarova G.I. Oxyproline content in the tissues was determined by
Bergman and Loksley method modified by Borisova L.B. with co-authors. The determination
of hexuronic acids was made according to the description of Altynbekov B.E.
with co-authors. Lipids content was determined by the turbidity method. The
statistical processing of the analyzed material was made according to the table
of (t) criteria values after Student.
In the result of the acute experiment it was established that after exposure to hexane, the TBARP content in spleen of rats increased by 34% immediately after exposure. Thereafter that value reduced. In 3 hours there was no significant difference from the reference level and from the value obtained immediately after exposure. In 12 hours there was noted significant difference in the TBARP level as compared with the reference group, the TBA-RP level immediately after exposure and the same in the 3 hours after exposure, by 20%, 18% and 16% respectively. The level of diene conjugates immediately after exposure and in 3 hours thereafter was the same as the reference one. However, in 12 hours it increased by 20% as compared with the reference level. Catalase activity level was not changed in actual fact in the course of the whole observation period. Content of Cholesterin, oxyproline and hexuronic acids in spleen was not changed.
Thus, the results of the acute experiment using the dose of 1/3 CL50 showed that in case of short-time exposure to hexane there is no impact upon the condition of erythrocytes. At that, in spleen there was stimulated cyclooxygenase oxidation of polysaturated fatty acids immediately after exposure to hexane which was demonstrated by the increase in TBA-RP level by 34%. However, already in 3 hours and at the end of the observation period that reaction diminished. At that, in 12 there was noted the intensification of leukotriene oxidation of polysaturated fatty acids in the form of increase in DC level as compared with all other groups. During the whole observation period there was no change in the level of antioxidant activity in spleen which is proved by the fact that there was no change in caltalase activity in spleen and no change in the levels of Cholesterin, oxyproline and hexuronic acids which is evidence of no change in the structure of the spleen tissues membranes and no change in the connective tissues metabolism in that organ.
